Description pSKI015 insertion. Arabidopsis Knockout Facility at the University of Wisconsin-Madison
|
Phenotype
Loss of JAGGED function causes organs to have serrated margins. In leaves, the blade region is most severely affected. In sepals, petals and stamens, the strongest defects are seen in the distal regions. By monitoring cell-cycle activity in developing petals with the expression of HISTONE 4, we show that JAGGED suppresses the premature differentiation of tissues, which is necessary for the formation of the distal region. The localization of defects overlaps with the expression domain of JAGGED, which is restricted to the growing regions of lateral organs.

Description Single basepair substitution mutant isolated from EMS-mutagenized Arabidopsis Col-gl1 ecotype, using the chlorophyll accumulation assay (Burke et al. Plant Physiol. 123, 575-588, 2000). The mutant was backcrossed to Col-gl wild type at least three times before genetic and physiological characterization. The recessive nature of the mutation was evaluated using F1 seedlings from a backcross, whereas the segregation ratios of mutant phenotype were assessed using F2 seedlings.

Description Transgenic line tagged with fluorescent proteins. Nuclei, plasma membranes, mitochondria, and plastids are genetically tagged with CFP, GFP, YFP, and RFP respectively. Agrobacterium tumefaciens strain GV3101/pMP90 was transformed with the vectors, pKW102-CY-MODC, pEL103-RecA::DsRed2 , and pEL103-GR respectively. These Agrobacterium strains were then used to transform Arabidopsis thaliana (Columbia accession). Zygosities of each transgenic line were determined by their mortalities on kanamycin selection plates (50 ug/ mL) and by microscopic detections of the fluorescent proteins in their seedlings. Homozygous lines of transgenic plants expressing Gal4-CFP and CoxIV-YFP were crossed to homozygous lines of transgenic plants expressing Cam53BD-GFP. The resulting homozygous lines were further crossed to homozygous transgenic plants expressing RecA-RFP to obtain the Kaleidocell lines. The line that most strongly expressed all transgenes was designated Kaleidocell.17. Kaleidocell.17 was self-pollinated in the following three generations. The line that expressed all transgenes in its all progenies was identified and designated Kaleidocell.17.1.7. The progenies of Kaleidocell.17.1.7 were used as the homozygous of Kaleidocell. The second progenies of Kaleidocell.17.1.7 (designated Kalediocell.17.1.7.7.X) are the distribution stock.

Phenotype
