Description

Moore Transactivation-enhancer Trap population

This population contains enhancer traps, generated with the enhancer-trap vector pHET-LhG4. They provide misexpression patterns for use in the pOp/LhG4 system.

The pOp/LhG4 system is used for spatial and temporal control of transgene expression in plants. This is based on a chimeric transcription factor, LhG4, comprising a high-affinity DNA-binding mutant of the E. coli lac repressor fused to a transcription activation domain from the yeast Gal4 protein. This molecule activates transcription from the pOp promoter which is otherwise physiologically silent in transgenic plants.

Stock description

Columbia plants were transformed with a pOpGUS reporter plasmid (Hygromycin resistant) as described in Rutherford et al. (2005). Some lines still segregate for the enhancer trap- can be selected on kanamycin. All lines are projeny from T2 plants described in Rutherford et al. (2005). In most cases 2 independent T3 familes are supplied designated by the letter b or t followed by a number. HET:50d, HET:80d, HET:82c-b2, HET:114y-HT1 and HET114y-HT3 were not described by Rutherford et al. (2005) but information about their expression patterns can be obtained from the University of Oxford link below (www.plants.ox.ac.uk).
